After Admitting Mistake, AstraZeneca Faces Difficult Questions About Its Vaccine

The announcement this week that a cheap, easy-to-make coronavirus vaccine appeared to be up to 90 percent effective was greeted with jubilation. But since unveiling the preliminary results, AstraZeneca has acknowledged a key mistake in the vaccine dosage received by some study participants, adding to questions about whether the vaccine’s apparently spectacular efficacy will hold up under additional testing. Scientists and industry experts said the error and a series of other irregularities and omissions in the way AstraZeneca initially disclosed the data have eroded their confidence in the reliability of the results. The regimen that appeared to be 90 percent effective was based on participants receiving a half dose of the vaccine followed a month later by a full dose; the less effective version involved a pair of full doses. AstraZeneca disclosed in its initial announcement that fewer than 2,800 participants received the smaller dosing regimen, compared with nearly 8,900 participants who received two full doses. Moncef Slaoui, the head of Operation Warp Speed, the U.S. initiative to fast-track coronavirus vaccines, noted another limitation in AstraZeneca’s data. On a call with reporters, he suggested that the participants who received the half-strength initial dose had been 55 years old or younger. The fact that the initial half-strength dose wasn’t tested in older participants, who are especially vulnerable to Covid-19, could undermine AstraZeneca’s case to regulators that the vaccine should be authorized for emergency use.

Government Model Suggests U.S. COVID-19 Cases Could Be Approaching 100 Million

The actual number of coronavirus infections in the U.S. reached nearly 53 million at the end of September and could be approaching 100 million now, according to a model developed by government researchers. The model, created by scientists at the Centers for Disease Control and Prevention, calculated that the true number of infections is about eight times the reported number, which includes only the cases confirmed by a laboratory test. Preliminary estimates using the model found that by the end of September, 52.9 million people had been infected, while the number of laboratory-confirmed infections was just 6.9 million, the team reported in the Nov. 25 issue of the journal Clinical Infectious Diseases. "This indicates that approximately 84% of the U.S. population has not yet been infected and thus most of the country remains at risk," the authors wrote. Since then, the CDC's tally of confirmed infections has increased to 12.5 million. So if the model's ratio still holds, the estimated total would now be greater than 95 million, leaving about 71% of the population uninfected. The model attempts to account for the fact that most cases of COVID-19 are mild or asymptomatic and go unreported. Scientists used studies looking for people who have antibodies to the coronavirus in their blood – an indication that they were infected at some time — to estimate how many infections went undetected. Some of these antibody studies have suggested that only about one in 10 coronavirus infections is reported.

Companies are launching unprecedented numbers of rockets to put global Wi-Fi and 5G satellites into the sky. 100,000+ satellites, plus drones and balloons, are planned. Industry and government have done little research, claiming the “big sky” can absorb any problems. And the FCC exempts these satellite networks from environmental review. The sun turns oxygen into ozone in the stratosphere, creating the ozone shield which protects the Earth. But rocket exhaust, alumina, water vapor, and black carbon accumulate in the stratosphere and block the sun’s rays, reducing ozone creation. In 2018, Martin Ross, senior project engineer at The Aerospace Corporation, estimated rockets’ black carbon and alumina was 11,000 tons per year. These new mega-constellations require 1000s of additional rocket launches. Severe ozone loss is possible. 253 scientists signed the 2015 International EMF Scientist Appeal, warning the UN and member states of the damage already occurring. Extensive research shows many impacts from this radiation including: increased tumor, cancer, and stroke risk, oxidative stress, increase in free radicals, DNA, neurological, immune, and dermatological damage, heart rhythm disorders ... cognitive problems, headaches, nausea, and links to Alzheimer’s and ADHD. Wildlife, bees, birds, trees, and plants are also harmed. 5G millimeter RF is highly absorbed by the skin’s sweat ducts and the cornea of the eye, and could additionally impact cardiac function.

Eriko Kobayashi has tried to kill herself four times. The first time, she was just 22 years old with a full-time job in publishing that didn't pay enough to cover her rent and grocery bills in Tokyo. "I was really poor," said Kobayashi, who spent three days unconscious in hospital after the incident. Now 43, Kobayashi has written books on her mental health struggles and has a steady job at an NGO. But the coronavirus is bringing back the stress she used to feel. "My salary was cut, and I cannot see the light at the end of the tunnel," she said. "I constantly feel a sense of crisis that I might fall back into poverty." Experts have warned that the pandemic could lead to a mental health crisis. Mass unemployment, social isolation, and anxiety are taking their toll on people globally. In Japan, government statistics show suicide claimed more lives in October than Covid-19 has over the entire year to date. The monthly number of Japanese suicides rose to 2,153 in October. As of Friday, Japan's total Covid-19 toll was 2,087, the health ministry said. Japan is one of the few major economies to disclose timely suicide data -- the most recent national data for the US, for example, is from 2018. The Japanese data could give other countries insights into the impact of pandemic measures on mental health. For the 10 years leading up to 2019, the number of suicides had been decreasing in Japan. The pandemic appears to have reversed that trend.

Purdue Pharma pleads guilty to federal criminal charges related to nation's opioid crisis

Purdue Pharma, the maker of OxyContin, pleaded guilty Tuesday to three federal criminal charges related to the company's role in creating the nation's opioid crisis. Purdue Pharma board chairman Steve Miller pleaded guilty on behalf of the company during a virtual federal court hearing in front of US District Judge Madeline Cox Arleo. The counts include one of dual-object conspiracy to defraud the United States and to violate the Food, Drug, and Cosmetic Act, and two counts of conspiracy to violate the Federal Anti-Kickback Statute. The plea deal announced in October includes the largest penalties ever levied against a pharmaceutical manufacturer, including a criminal fine of $3.544 billion and an additional $2 billion in criminal forfeiture, according to a Department of Justice press release. According to the US Centers for Disease Control and Prevention, about 70,000 Americans died of drug overdoses in 2018, just one year of the opioid crisis, and about 70% of those deaths were caused by prescription or illicit opioids like OxyContin. Several civil lawsuits against Purdue Pharma related to the opioid crisis are still ongoing as the company undergoes bankruptcy proceedings. The Plaintiffs' Executive Committee in the National Prescription Opiate Litigation Multi-District Litigation called Purdue Pharma's guilty plea "long overdue." "Their illegal and profit-seeking actions were egregious. It is important to note, however, that they are just one company in one part of the larger opioid supply chain," [the plaintiffs'] attorneys ... said.

Evidence of 90-million-year-old rainforest uncovered beneath Antarctic ice

Antarctica is now a harsh land of ice and snow, but has not always been that way. Earth’s southernmost continent long ago was home to temperate, swampy rainforests teeming with life, scientists said on Wednesday based on pristinely preserved forest soil they retrieved by drilling under the seafloor off Antarctica’s coast. The sediment core obtained by scientists working aboard the research icebreaker RV Polarstern in the Amundsen Sea near the Pine Island Glacier dated to about 90 million years ago during the Cretaceous Period when dinosaurs were the dominant land animals. The researchers estimated based on the soil content that this location, 560 miles (900 km) from the South Pole, experienced average annual temperatures of about 53-55 degrees Fahrenheit (12-13 Celsius) and average temperatures during the warmest summer months of about 68-77 Fahrenheit (20-25 Celsius). A modern temperature analogue may be New York City, according to marine geologist Johann Klages ... lead author of the research published in the journal Nature. The dark-brownish gray soil was composed of fine-grained silt and clay bearing remains of fossil roots in a dense network, pollen and spores spanning 65 types of plants, with individual cell structures clearly visible. “If you would go to a forest near you and drill a hole, it would probably look pretty similar,” Klages said. While they did not find any animal remains, Klages said there likely were dinosaurs.

For the first time, the number of children paralyzed by mutant strains of the polio vaccine are greater than the number of children paralyzed by polio itself. So far in 2017, there have been only six cases of "wild" polio reported anywhere in the world. By "wild," public health officials mean the disease caused by polio virus found naturally in the environment. By contrast, there have been 21 cases of vaccine-derived polio this year. These cases look remarkably similar to regular polio. But laboratory tests show they're caused by remnants of the oral polio vaccine that have gotten loose in the environment, mutated and regained their ability to paralyze unvaccinated children. The oral polio vaccine used throughout most of the developing world contains a form of the virus that has been weakened in the laboratory. But it's still a live virus. (This is a different vaccine than the injectable one used in the U.S. and most developed countries. The injectable vaccine is far more expensive and does not contain live forms of the virus.) WHO is attempting to phase out the use of live oral polio vaccine to eliminate the risk that the active virus in the vaccine could mutate into a form that can harm unvaccinated children. But for now, the live vaccine continues to be the workhorse of the global polio eradication campaign. The killed vaccine doesn't fully block the virus from spreading because a person who is immunized can still carry and spread the polio virus.

Is the FDA Too Cozy With Drug Companies?

People who work at the U.S. Food and Drug Administration (FDA) as medical reviewers are responsible for parsing the risks and benefits of a particular drug before it gets the agency’s approval. But a new report from two researchers at the Oregon Health and Science University, published in the journal The BMJ, suggests many of these medical reviewers go on to work for the drug companies they oversaw while working for the government. The study’s authors ... looked at the FDA’s list of haematology-oncology drug approvals from 2006 to 2010 and scanned all medical reviews from 2001 to 2010 in the agency’s database, then looked up the subsequent jobs of the people who worked as medical reviewers for those drug approvals. The researchers found that among 55 people who worked as haematology-oncology medical reviewers from 2001 to 2010, 27 continued in their roles at the FDA, two people worked at the FDA but held other appointments, and 15 left the FDA to work with or consult for the biopharmaceutical industry. “If you know in the back of your mind that a major career opportunity after the FDA is going to work on the other side of the table, I worry it can make you less likely to put your foot down,” says study author Dr. Vinay Prasad. “Regulators may be less willing to be very tough, and I worry that is happening.” Prasad says he would like to see more transparency from the FDA on the number of people who go from the agency to the drug industry.

Those with the gold make the rules. In an investigative series [the Miami Herald] documents the whitewash of an alleged global conspiracy to traffic underage girls for sexual exploitation. Though [the Herald] identified more than 80 likely victims, [Jeffrey Epstein] was allowed – under a furtive plea deal – to serve just 13 months in country-club conditions. Epstein, a fantastically wealthy creep, ran afoul of the Palm Beach police in 2005 after the parents of a 14-year-old girl reported that he paid their daughter to strip and massage his naked body while he pleasured himself. Investigators soon found evidence ... indicating that troubled girls by the dozens were recruited for molestation and rape. Epstein’s wealth – the origins of which are a bit murky – assembled an all-star team of defense lawyers, including Alan Dershowitz, the Harvard scholar. Left unresolved is whether Epstein’s extensive array of powerful friends may have helped him out, too. In the same “little black book” where he kept the names of underage girls around the world available for “massage,” Epstein also had contact information for Donald Trump, Bill Clinton, Michael R. Bloomberg, Prince Andrew, assorted Kennedys and so on. Some of Epstein’s accusers [have been silenced]. Silent, too, is Trump, who once claimed a 15-year acquaintance with Epstein, whom he described as “a lot of fun.” Trump noted Epstein’s interest in women “on the younger side.” And Clinton is uncharacteristically mute, though he used to spend so much time on Epstein’s private jet – dubbed “the Lolita Express” – that, if it were an airline, he’d have platinum status. Two of our past four presidents have been chummy with a registered sex offender. Oslo got pedestrian and cyclist deaths down to zero. Here’s how

In 2019, Oslo, Norway recorded zero pedestrian or cyclist deaths. There was only a single traffic fatality, which involved someone driving into a fence. (For comparison, preliminary figures in London show 73 pedestrian and six cyclist fatalities in 2019; New York recorded 218 total traffic fatalities, including 121 pedestrian and 28 cyclist deaths.) Oslo’s achievement means that it is just one step away from “Vision Zero”, an undertaking to eliminate all deaths on public roads. The foundation for reaching Vision Zero is to significantly reduce the number of cars on the road. Oslo officials have removed more than a thousand street-side central parking spots, encouraging people to lean on an affordable and flexible public transport network, and added more bike lanes and footpaths. Significant areas are closed off to cars entirely, including “heart zones” around primary schools. “The wish to pedestrianise the city isn’t a new policy, but it has accelerated now,” Rune Gjøs, a director at Oslo’s Department of Mobility, says. “The city centre is now a thriving area and all the top-brand shops want to establish themselves on the car-free streets,” Gjøs says. “This shows that consumers find these streets attractive, and they’re leaving as much money behind as if they were coming by car.” Demand for residential real estate has also increased, thanks to lower levels of traffic and pollution.

Antibodies Good. Machine-Made Molecules Better?

The coronavirus might be new, but nature long ago gave humans the tools to recognize it, at least on a microscopic scale: antibodies, Y-shaped immune proteins that can latch onto pathogens and block them from infiltrating cells. Millions of years of evolution have honed these proteins into the disease-fighting weapons they are today. But in a span of just months, a combination of human and machine intelligence may have beaten Mother Nature at her own game. Using computational tools, a team of researchers at the University of Washington designed and built from scratch a molecule that, when pitted against the coronavirus in the lab, can attack and sequester it at least as well as an antibody does. This molecule, called a mini-binder for its ability to glom onto the coronavirus, is petite and stable enough to be shipped en masse in a freeze-dried state. Bacteria can also be engineered to churn out these mini-binders, potentially making them not only effective but also cheap and convenient. Eventually, healthy people might be able to self-administer the mini-binders as a nasal spray, and potentially keep any inbound coronavirus particles at bay. Mini-binders are not antibodies, but they thwart the virus in broadly similar ways. The coronavirus enters a cell using a kind of lock-and-key interaction, fitting a protein called a spike — the key — into a molecular lock called ACE-2, which adorns the outsides of certain human cells. Antibodies made by the human immune system can interfere with this process.

Couple uses catering deposit from canceled wedding to feed 200 people on Thanksgiving

Like many weddings this year, Emily Bugg and Billy Lewis' nuptials didn't go as planned. Because of coronavirus restrictions, the couple decided to get married at City Hall in Chicago instead of having a big ceremony. And instead of taking the deposits for their reception back, they decided to repurpose them. The couple put their $5,000 worth of reception food to a good use on Thanksgiving, according to a local charity. Bugg and Lewis donated the 200 meals to Thresholds, an organization that provides services and resources for people with serious mental illnesses and substance use disorders in Illinois. Thresholds usually holds a communal Thanksgiving dinner for clients, but it was canceled due to COVID-19 gathering restrictions. Instead, Bugg and Lewis' wedding caterer, Big Delicious Planet, put the couple's $5,000 deposit to use to prepare special Thanksgiving meals for delivery. The caterers worked alongside Threshold staff members to box individual meals, which where then delivered to the client's homes. Big Delicious Planet cooked turkey, dressing, mashed potatoes, green beans and other Thanksgiving staples. "Canceling a big wedding isn't the worst thing that could happen," Bugg said. "We're happy to be married, and we're so happy that we could help Thresholds' clients ... as a result of the wedding cancellation." Thresholds CEO Mark Ishaug said the couple's donation is "an incredible example of the generosity and creativity that the pandemic has inspired in so many."

Dolphins Guide Scientists to Rescue Suicidal Girl

My research team and I were following a school of bottlenose dolphins near shore ... off Los Angeles, California. The dolphins were still feeding in circle near shore, when suddenly, one individual changed direction heading out toward deeper water. A minute later, the rest of the school turned to follow. Seeing them abruptly leave a foraging ground and change direction came as a surprise to the research team. I decided to follow them. The dolphins increased their speed. Somewhere near three miles offshore the dolphin group stopped, forming a sort of ring around a dark object in the water. “Someone’s in the water!” yelled my assistant, standing up and pointing at the seemingly lifeless body of a girl. As the boat neared, she feebly turned her head toward us, half-raising her hand as a weak sign for help. If we didn’t act immediately, the girl would die. We [pulled] the frail and hypothermic body on board. “She is cyanotic,” said one of my researchers, also a lifeguard, after a cursory examination. “She has severe hypothermia. We need to get her warm!” We managed to get some of her wet garments off and wrap her in a blanket. We took turns keeping her warm by huddling with her under the blanket. A couple of hours later, we were all waiting outside the emergency room at the Marina del Rey hospital. The ER doctor came out to talk with us. The girl, it seems, would pull through, and he thanked us for our quick action. He tells us the girl was vacationing in L.A. from Germany and, as the letter found in her plastic bag explained, she was attempting suicide. If we hadn’t found her, if the dolphins hadn’t led us offshore when they did, to that specific place, she would have died.
